Duringthelaterpartofthe20thcentury,pharmaceuticalcompoundscontain-
ingmetalsbegantoplayanincreasinglyimportantroˆleinmedicine.In
particular,thediscoveryofplatinumanticancerdrugsandtheappearanceof
organspecificdiagnostic-imagingagentscontainingtechnetiumwerepromi-
nentdevelopmentsthathavestimulatedfurtherinterestinso-calledmetallo-
pharmaceuticals.Today,themetallopharmaceuticalsindustryhasaglobal
marketmeasuredinbillionsofpoundsandhaswell-establishedapplicationsin
bothdiagnosticandtherapeuticmedicine.
Theaimofthisbookistoreviewthediagnosticandtherapeuticapplications
ofselectedmetallicelementsandconsiderthechemistryunderlyingtheformu-
lationofmetallopharmaceuticals.Inordertodothisitisnecessarytoassume
somebasicunderstandingofthechemicalbehaviourofmetalsand,inpartic-
ular,theircoordinationchemistry.Averybriefintroductiontocoordination
chemistryisalsopresentedforthebenefitofthoseunfamiliarwiththisaspectof
chemistry.Thelevelofpresentationofthissectionisdesignedforageneralbut
scientificallyliterateaudience,someofwhommayhaveonlyarudimentary
knowledgeofchemistry.Itishopedthetextwillbehelpfultopharmacistsas
wellastocliniciansandmedicalresearchersusingmetalcontainingdrugsina
clinicalenvironment.However,itisalsohopedthatthebookmightalsoserve
asanintroductorytextofinteresttochemistrystudentswishingtolearnabout
anaspectoftheirsubjectwithinanappliedcontext,ratherthanjustasasubject
initsownright.
Thediscussionofmedicinalapplicationsfocusesonthecoordinationchem-
istryofwhatareknownasthetransitionalelements,particularlyvanadium,
manganese,technetium,rutheniumrhodium,platinum,goldandgadolinium.
Compoundscontainingmetalsofthistypearethemostamenabletomanip-
ulationthroughchemicaldesignsothatcoordinationchemistryplaysasignif-
icantroˆleinthedevelopmentofmetallopharmaceuticalsinvolvingthesemetals.
Theso-calledmaingroupmetals(e.g.gallium,indium,tin,lead,bismuth)are,
forthemostpart,notincludedthoughsomeoftheirapplicationsaremen-
tioned.Theseelementsofferlessscopefortheapplicationofcoordination
chemistryinproductdevelopmentastheircompoundstypicallybreakdown
quicklyinvivotransferringthemetaltobiologicalbindingagentsandafate
largelybeyondthechemistsdirectcontrol.
Thefirstintroductorychapteraimstosetthescenewithabriefhistorical
reviewandaconsiderationofsomegeneralaspectsoftheuseofchemical
